چکیده
Microbial metabolic pathway engineering is a potent strategy used worldwide to produce aromatic compounds. We drastically rewired the primary of Escherichia coli aromatics and their derivatives. The E. was compartmentalized into production energy modules. focused on pyruvate-forming reaction in biosynthesis some compounds as connecting those strains were engineered show no growth unless pyruvate synthesized along with interest production. Production salicylate maleate demonstrated confirm our strategy's versatility. In production, yield against theoretical yield, rate reached 12.0 g L?1, 67%, up fourfold compared that previous reports, respectively; these are highest values microbes knowledge. results reveal strongly promotes

The mechanism of catalysis of the chorismate to prephenate reaction by the Escherichia coli mutase enzyme.
Molecular dynamics studies of the Escherichia coli chorismate mutase (EcCM), containing at the active site chorismate and in turn the transition state (TS), have been performed. The simulations show that TS is not bound any tighter than chorismate. Comparison of average polar interactions show they are virtually identical for interactions of EcCM with chorismate and the TS, whereas hydrophobic ...
متن کاملFormate fixation in pyruvate by Escherichia coli.
The phosphoroclastic decomposition of pyruvic acid to acetic and formic acids by enzyme extracts of Escherichia coli has been an object of study ever since acetyl phosphate was tentatively identified as an intermediate of this reaction (1). The finding by Utter, Lipmann, and Werkman that isotopic carbon of formate was incorporated into the carboxyl group and that the carboxyl carbon of acetate ...
متن کاملExpression of an anaplerotic enzyme, pyruvate carboxylase, improves recombinant protein production in Escherichia coli.
Anaplerotic enzyme reactions are those which replenish tricarboxylic acid intermediates that are withdrawn for the synthesis of biomass. In this study, we examined recombinant protein production in Escherichia coli containing activity in an additional anaplerotic enzyme, pyruvate carboxylase. In batch fermentations, the presence of pyruvate carboxylase resulted in 68% greater production of the ...
متن کاملModeling of the pyruvate production with Escherichia coli: comparison of mechanistic and neural networks-based models.
Three different models: the unstructured mechanistic black-box model, the input-output neural network-based model and the externally recurrent neural network model were used to describe the pyruvate production process from glucose and acetate using the genetically modified Escherichia coli YYC202 ldhA::Kan strain. The experimental data were used from the recently described batch and fed-batch e...
متن کاملProduction of MAG1 Antigen of Toxoplasma gondii in Escherichia coli
Introduction : Toxoplasmosis is a parasitic infection caused by the protozoan Toxoplasma gondii it leads to serious medical problems in congenitally-infected and immunocompromised individuals, while it is quite harmless in immunocompetent individuals. Toxoplasma tissue cyst matrix protein (MAG1) induces early humoral and cell-mediated immune responses. Previous studies suggested recombinant M...
